Fish-Pathogen challenge protocol

I am doing a transcriptomic study on fish immunogenomics, which involves fish-pathogen challenge. To do this I would like to know the parameters that I need consider. Especially, what is the appropriate concentration of bacteria to be used? Is strain-specific or fish species specific or both.
For additional info I am will be working on Lates Calcarifer challenged with live V. alginolyticus. But I do not know what concentration to use, is it dependant on LD-50?
